Blocking Translation of Oncogenic mRNA

چکیده

Double-stranded RNA-mediated interference (RNAi), antisense oligonucleotides (ASO), and ribozymes have excellent specificity to their target oncogenic mRNA. They also seem show great promise when it comes treating cancer. The problem is that RNAi, ASO, poor stability are constantly being degraded by nucleases. Researchers made some efforts increase oligonucleotides’ creating phospharimidate Phosphorothioate. Currently, ribozymes, oligonucleotides, (RNAi) the three main methods used RNA. These currently undergoing clinical trials for purpose of focusing on specific RNAs involved in disorders like cancer neurodegeneration. In fact, ASOs amyotrophic lateral sclerosis spinal muscular atrophy produced promising results trials. formation chemical alterations boost affinity selectivity while reducing noxiousness owing off-target impacts two benefits ASOs. Another benefit increased affinity. With a focus RNAi ASOs, this review illustrated therapeutic strategies RNA therapy now use.

Blocking oncogenic Ras signaling for cancer therapy.

The Ras gene product is a monomeric membrane-localized G protein of 21 kd that functions as a molecular switch linking receptor and nonreceptor tyrosine kinase activation to downstream cytoplasmic or nuclear events. Each mammalian cell contains at least three distinct ras proto-oncogenes encoding closely related, but distinct proteins. Activating mutations in these Ras proteins result in consti...

A MAPK/HNRPK pathway controls BCR/ABL oncogenic potential by regulating MYC mRNA translation.

Altered mRNA translation is one of the effects exerted by the BCR/ABL oncoprotein in the blast crisis phase of chronic myelogenous leukemia (CML). Here, we report that in BCR/ABL+ cell lines and in patient-derived CML blast crisis mononuclear and CD34+ cells, p210(BCR/ABL) increases expression and activity of the transcriptional-inducer and translational-regulator heterogeneous nuclear ribonucl...

Sensitivity of mRNA Translation

Using the dynamic mean-field approximation of the totally asymmetric simple exclusion process (TASEP), we investigate the effect of small changes in the initiation, elongation, and termination rates along the mRNA strand on the steady-state protein translation rate. We show that the sensitivity of mRNA translation is equal to the sensitivity of the maximal eigenvalue of a symmetric, nonnegative...

Regulation of mRNA translation.

The control of mRNA translation plays an important role in regulating gene expression in diverse situations. Nutrients, especially amino acids, regulate translation to control the expression of specific proteins including transcriptional activators and ribosomal proteins. Sequence elements in the 5'- or 3'-untranslated regions of mRNAs can control their translation. Hormones such as insulin act...

Controlling mRNA Translation.

The ribosomal density along different parts of the coding regions of the mRNA molecule affects various fundamental intracellular phenomena including: protein production rates, global ribosome allocation and organismal fitness, ribosomal drop off, co-translational protein folding, mRNA degradation, and more. Thus, regulating translation in order to obtain a desired ribosomal profile along the mR...
